A RelC deletion mutant, KO-100, of Streptomyces coelicolor A3(2) has been isolated from a collection of spontaneous thiostrepton-resistant mutants. KO-100 grows as vigorously as the parent strain and possesses a 6-bp deletion within the rplK, previously termed relC. When the wild-type rplK gene was propagated on a low-copy-number vector in mutant KO-100, the ability to produce ppGpp, actinorhodin and undecylprodigiosin, which had been lost in the RelC mutant, was completely restored. Allele replacement by gene homogenotization demonstrated that the RelC mutation is responsible for the resistance to thiostrepton and the inactivation of ppGpp, actinorhodin and undecylprodigiosin production. Western blotting showed that ribosomes from the RelC mutant KO-100 contain only one-eighth the amount of L11 protein found in ribosomes of the parent strain. The impairment of antibiotic production in KO-100 could be rescued by the introduction of mutations that confer resistance to streptomycin (str), which result in alteration of Lys-88 in ribosomal protein S12 to Glu or Arg. Nanoflow electrospray mass spectrometry was used to monitor the formation of protein heterodimers of HU proteins from Bacillus stearothermophilus and Bacillus subtilis. This has enabled us to analyze both thermodynamic and kinetic features associated with the dissociation of homodimeric HU proteins. The results obtained correlate well with the kinetics of the protein dissociation process and the free energy difference between homo- and heterodimeric species anticipated from other studies. We suggest that this approach will have general applicability in studying protein association and dissociation under near-equilibrium conditions and will be relevant to a wide range of biological systems.  相似文献

Habitat fragmentation and modification have been recognized as the most serious threats to biodiversity. In Croatia, the intensive human activities resulted in the landscape transformation, reduction of grassland areas and loss of species number. Increasingly threatened are the wild edible and medicinal plants which contribute remarkably to the biodiversity of different grassland types. In northeastern Croatia, one of the last remnants of the steppe-like grassland remained preserved in a very complex anthropogenic landscape. We examined the diversity of vascular plants occurring on this small habitat patch. Among the relatively high number of plant taxa (in a total of 177) found during the two years of investigations, most could be used both for human nutrition (28 taxa), and for the treatment of a wide range of diseases (60 taxa), while some taxa have nutritional value for livestock (17 taxa). All 23 recorded taxa with edible and medicinal properties are threatened in Croatia, 21 are threatened across Europe, and two are Red Listed taxa. Our results showed that small and isolated steppe-like grassland represents an important refuge for variety of valuable plant species in intensively used landscapes. Accordingly, such small habitats should be, at least locally, effectively protected.  相似文献

Cryopreserved canine adipose-derived mesenchymal stromal cells (Ad-MSCs) can be used instantly in dogs for clinical uses. However, cryopreservation results in a reduction of the cellular viability, proliferation, and anti-oxidation of post-thawed Ad-MSCs. Therefore, there is a need for in-vitro procedure to improve post-thawed Ad-MSCs’ viability, proliferation, anti-oxidation, and differentiation capacity. In this study, fresh-Ad-MSCs were activated with heat shock, hypoxia (5% O2), or hypoxia (5% O2) + heat shock treatments. The results showed that compared to the other treatments, heat shock significantly improved the proliferation rate, anti-oxidation, heat shock proteins and growth factors expressions of canine-fresh-Ad-MSCs. Consequently, fresh-Ad-MSCs were heat-shocked and then cryopreserved with different combinations of dimethyl sulfoxide (Me2SO) and fetal bovine serum (FBS) to determine the combination that could effectively preserve the cellular viability, proliferation, anti-oxidation and differentiation capacity of Ad-MSCs after cryopreservation. We found that C-HST-Ad-MSCs cryopreserved with 10% Me2SO + 40% FBS presented significantly (p < 0.05) improved cellular viability, proliferation rate, anti-oxidant capacity, and differentiation potential as compared to C-HST-Ad-MSCs cryopreserved with 1% Me2SO + 10% FBS or 1% Me2SO alone or control. We concluded, heat shock treatment is much better to enhance the characteristics of fresh-Ad-MSCs than other treatments, moreover, C-HST-Ad-MSCs in 10% Me2SO + 40% FBS showed better results compared to other cryopreserved groups. However, future work is required to optimize the expression of heat shock proteins, which would further improve the characteristics of fresh- and cryopreserved-HST-Ad-MSCs and reduce the dependency on Me2SO and FBS.  相似文献

MYCN amplification is an independent risk factor for poor prognosis in neuroblastoma (NB), but its protein product cannot be directly targeted because of protein structure. Thus, this study aimed to explore novel ways to indirectly target N-Myc by regulating its post-translational modifications (PTMs) and therefore protein stability. N-Myc coimmunoprecipitation combined with HPLC–MS/MS identified 16 PTM residues and 114 potential N-Myc-interacting proteins. Notably, both acetylation and ubiquitination were identified on lysine 199 of N-Myc. We then discovered that p300, which can interact with N-Myc, modulated the protein stability of N-Myc in MYCN-amplified NB cell lines and simultaneously regulated the acetylation level and ubiquitination level on lysine-199 of N-Myc protein in vitro. Furthermore, p300 correlated with poor prognosis in NB patients. Taken together, p300 can be considered as a potential therapeutic target to treat MYCN-amplified NB patients, and other identified PTMs and interacting proteins also provide potential targets for further study.  相似文献

Selection programs have enabled broiler chickens to gain muscle mass without similar enlargement of the cardiovascular and respiratory systems that are essential for thermoregulatory efficiency. Meat-type chickens cope with high ambient temperature by reducing feed intake and growth during chronic and moderate heat exposure. In case of acute heat exposure, a dramatic increase in morbidity and mortality can occur. In order to alleviate heat stress in the long term, research has recently focused on early thermal manipulation. Aimed at stimulation of long-term thermotolerance, the thermal manipulation of embryos is a method based on fine tuning of incubation conditions, taking into account the level and duration of increases in temperature and relative humidity during a critical period of embryogenesis. The consequences of thermal manipulation on the performance and meat quality of broiler chickens have been explored to ensure the potential application of this strategy. The physiological basis of the method is the induction of epigenetic and metabolic mechanisms that control body temperature in the long term. Early thermal manipulation can enhance poultry resistance to environmental changes without much effect on growth performance. This review presents the main strategies of early heat exposure and the physiological concepts on which these methods were based. The cellular mechanisms potentially underlying the adaptive response are discussed as well as the potential interest of thermal manipulation of embryos for poultry production.  相似文献

The aim of the work was to elucidate the presence of different hydrogen bond (H-bond) in five Zif268 proteins (1A1F, 1A1G, 1A1H, 1A1I and 1A1K). For this purpose, we have performed the QM/MM and molecular dynamics (MD) studies, the results of which reveal that H-bonds depend on the amino acid sequence and orientation of the H-bond donor atoms. Further, high specificity of Arg and Asn is observed for guanine and adenine, respectively. Furthermore, both conventional and non-conventional hydrogen bond also exists in the proteins, among them N–H?O H-bonds are the strongest. Besides, the non-conventional bonds play a role in the protein folding and DNA stacking. From the QSAR properties, amino acids such as asparagine and aspartic acids are the major reactive sites in the Zif268 protein. The electron affinities of Zif268 proteins are high, so the charge transfer occurs from the DNA to the protein molecules. NBO analysis indicates the majority of charge transfer occurs from DNA to the corresponding anti-bonding orbital of the peptides. Root mean square deviation and Rg (radius of gyration) show that 1A1F is more compact and in native state during MD simulation. The minimum Rg leads to the large number of hydrogen bonds formation in 1A1F. Higher solvent accessible surface area in 1A1I indicates that the cavity inside the protein is large.  相似文献

Morphogenesis of the ciliate cortex has been viewed as an attractive model system for studying the mechanisms behind the ordered assembly of subcellular structure. Based on the assumption that identifying protein components of the cortex would facilitate the study of cortical assembly, I have produced a number of monoclonal antibodies directed against components of the cortex of Euplotes aediculatus. Several of these antibodies react with the proteins comprising the alveolar plates. These thin polygonal scales, each enclosed within a flattened membranous sac (alveolus) just beneath the cell membrane, tightly abut in a confluent monolayer that appears to lend form and rigidity to the Euplotes cell cortex. Reactivity and specificity of these monoclonal antibodies for the alveolar plates was shown by immunofluorescence staining of whole-cell preparations and of cryosections, and by immuno-gold staining of thin sections by electron microscopy. On immunoblots of SDS-PAGE separated whole-cell extracts, the plate proteins are revealed as two to three closely spaced bands centered at an Mr of 97 kDa, and a larger relative at 125 kDa. Comparative peptide mapping reveals that the members of the 97-kDa protein cluster are closely related. However, the 125-kDa polypeptide varies significantly from the 97-kDa members, and hence is not likely a synthetic precursor. Because bands of these Mr values are prominent in Coomassie blue-stained gels of whole-cell extracts, and are greatly enriched in purified cortical preparations, they likely represent the major proteins comprising the alveolar plates of E. aediculatus. Cell volumes and intracellular concentrations of major solutes of Chlorococcum submarinum were determined before and after salinity shocks. Cells were found to shrink in size by about 30% following changes from 0.1 to 0.5 M NaCl, there was a transitory increase in sodium concentration and more permanent increases in concentrations of potassium, proline and glycerol (the major osmolyte). Conversely, cells doubled in size after the reciprocal downshock, there was rapid loss of about 70% of the cells' glycerol to the medium, a much smaller loss of cellular potassium and a steady disappearance of proline from the cells. The respiratory and photosynthetic responses to salinity fluctuations were also studied. Salinity downshocks stimulated respiration by 30% and inhibited photosynthesis by 16% within 5 min, but within 2 h these rates were identical to control rates. Upshocks caused a slight inhibition of respiration, but decreased photosynthesis by 40% within 5 min and recovery took 2 h. Downshocks had little effect on chlorophyll fluorescence, however, Fo strongly increased and both Fm and Fv/Fm declined within 5 min of salinity increases. This is consistent with a decrease in efficiency of PS2.
